Product Specific Information

The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by thymocytes, subsets of mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and is critical for T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤0.25 µg/test (100 µL final volume). Optimal titration recommended.

Fluorophore Information:
PE-eFluor® 610 is excited by 488–561 nm lasers and emits at 607 nm. Recommended detection filter: 610/20 band pass (equivalent to PE-Texas Red®). Protect from light due to photo-induced oxidation.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) with minimal impact on brightness and compensation. Clone-specific performance should be determined empirically.

Target Information

CD4 is a co-receptor for MHC class II molecules and a receptor for HIV. It is expressed mainly on T helper cells, as well as monocytes, macrophages, and dendritic cells. CD4 is found in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues such as brain and gut. It plays a role in T-cell activation through association with the T-cell receptor complex and Lck kinase, and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
