Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by thymocytes, T helper cells, and monocytes. CD4 acts as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Applications Reported:
For flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.125 µg)/test in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test.

Fluorophore Information:
PE-eFluor® 610 excitation: 488–561 nm; emission: 607 nm. Recommended filter: 610/20 band pass (PE-Texas Red® equivalent). Ensure instrument compatibility.

Light Sensitivity:
Protect from light; sensitive to photo-induced oxidation.

Fixation:
Samples can be stored up to 3 days in dark at 4°C using IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) with minimal impact on brightness and FRET efficiency.

Laser Compatibility:
Excitation: 488–561 nm; Emission: 607 nm; Laser: Blue, Green, Yellow-Green.

Target Information

CD4 antigen is a co-receptor for HIV and interacts with MHC class II molecules. Expressed primarily on T helper cells, and also on monocytes, macrophages, dendritic cells. Found in thymus, lymph nodes, tonsils, spleen, and some non-lymphoid tissues. CD4 assists early T-cell activation via association with TCR complex and Lck kinase. May mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts identified (RefSeq, July 2017).
