Target Information

Vacuolar-type H⁺-ATPase (V-ATPase) is a multisubunit enzyme responsible for acidifying eukaryotic intracellular organelles. It pumps protons against an electrochemical gradient to synthesize ATP.
V-ATPase consists of:

  • V1 domain: responsible for ATP hydrolysis (subunits A–H)
  • V0 domain: responsible for proton translocation (subunits a, d, c, c′, c″)

ATP6AP1 (ATPase, H⁺ transporting, lysosomal accessory protein 1) — also known as 16A, CF2, Ac45, XAP3, ATP6S1, VATPS1, or ATP6IP1 — is a type I transmembrane accessory protein of V-ATPase, predominantly expressed in endocrine and neuronal cells.
ATP6AP1 targets the V-ATPase enzyme to specialized vacuolar systems and interacts with V0 domain subunits via its cytoplasmic tail. Disruption of this interaction in osteoclasts results in impaired bone resorption, indicating its crucial role in osteoclastic function.
