SIK1은 LKB1의 생리적 효과(종양 억제 기능 포함)를 매개할 수 있으며, STRAD, NUAK1, NUAK2, BRSK1, BRSK2, QIK, QSK, SIK, MARK1, MARK2, MARK3, MARK4, MELK 등 다양한 AMPK 관련 단백질 신호 전달에 관여합니다.
Thr322 위치에서의 인산화에 의해 SIK1이 활성화되면 세포막의 Na⁺/K⁺ ATPase α-서브유닛의 촉매 활성이 증가하여 포유류 세포 내 나트륨 농도가 상승합니다.
이 단백질 계열은 티로신(PTK) 또는 세린/트레오닌(STK) 키나아제 촉매 도메인의 서열 비교를 기반으로 8개의 주요 그룹으로 분류됩니다.
STE 그룹(효모 Sterile 7, 11, 20 키나제의 상동체)은 MAPK 캐스케이드(MAPK, MAP2K, MAP3K, MAP4K)와 관련된 50개의 키나제로 구성됩니다.
MAP kinase 캐스케이드는 효모의 페로몬 반응 경로에서 가장 잘 연구되어 있으며, 페로몬이 Ste 세포 표면 수용체에 결합하여 MAPK 경로를 활성화합니다.
