For immunohistochemistry (PFA-fixed tissues):

• Incubate antibody for 30 minutes at room temperature (RT).
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at RT for 20 minutes.

Target Information

Desmoglein 3 (Dsg3), also known as Cadherin family member 6 (CDHF6), is part of the desmosomal cadherin family and plays a key role in cell-cell adhesion. It is predominantly expressed in stratified squamous epithelium, such as skin, oral mucosa, and esophagus.
Dsg3 is identified as the autoantigen in pemphigus vulgaris (PV), a severe autoimmune disease. Binding of anti-Dsg3 autoantibodies leads to loss of cell adhesion, resulting in blister formation on skin and mucosa and histologically showing suprabasilar acantholysis.
