Target Information

The TEK receptor tyrosine kinase (also known as TIE2) is expressed almost exclusively in endothelial cells. TEK signaling is crucial for endothelial cell–smooth muscle cell communication in venous morphogenesis, and defects in TEK are associated with inherited venous malformations. The ligand angiopoietin-1 binds to this receptor and mediates a signaling pathway involved in embryonic vascular development. Immunoblotting studies have shown that TIE2 expression increases in response to thyroid-stimulating hormone and agents that elevate intracellular cAMP. Hematopoietic stem cells (HSCs) expressing TIE2 are quiescent, antiapoptotic, and form a side population that adheres to osteoblasts within the bone marrow niche.
