Product Specific Information

Description:
The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta, is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Binding of 145-2C11 to TCR can initiate activation, proliferation, or apoptosis depending on conditions. It is widely used as a phenotypic marker for mouse T cells.

Applications Reported:
Used in flow cytometry, immunoprecipitation, immunoblotting (WB), and IHC of frozen tissues. Also utilized for cell activation and depletion. For functional assays, use Functional Grade purified 145-2C11 (Product #16-0031).

Applications Tested:
Validated in flow cytometric analysis of mouse thymocytes and splenocytes. Recommended ≤0.5 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. CD3 subunits (γ, δ, ε, ζ) are immunoglobulin superfamily members encoded on chromosome 11. They have cytoplasmic tails with tyrosine-based motifs mediating signal transduction. CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells. Not expressed on B or NK cells. CD3 gene defects are associated with immunodeficiency.
