Target Information
Adenoviruses are DNA viruses commonly found in nature and frequently cause acute upper respiratory tract infections such as common colds. There are 49 known human adenovirus serotypes divided into six subgroups (A–E). They are associated with respiratory illness, conjunctivitis, gastroenteritis, and may cause sexually transmitted diseases. Some types possess oncogenic potential, though most cause self-limiting febrile illnesses involving inflammation of conjunctivae and the respiratory tract.
Adenoviruses can persist latently in tissues such as tonsils and adenoids. In immunosuppressed patients (e.g., AIDS), reactivation may occur. Adenovirus vectors, particularly Ad5-based, are widely used in gene therapy research.
The early region (E1A) of the adenovirus genome plays a central role in cellular transformation and gene expression regulation. E1A encodes proteins (35–46 kDa) that interact with the retinoblastoma (Rb) tumor suppressor protein. Together with E1B, E1A forms the transforming region of adenovirus. E1A alone can immortalize primary cells, while E1B expression enables full transformation. Conserved regions of E1A share similarities with viral oncoproteins such as HPV-16/18 E7 and SV40 large T antigen.
