Product Specific Information

Description:
The M1/70 monoclonal antibody reacts with mouse CD11b, a 165–170 kDa integrin alphaM. CD11b non-covalently associates with CD18 to form the alphaMbeta2 integrin (Mac-1) and binds CD54 (ICAM-1), C3bi, and fibrinogen. Mac-1 is expressed by macrophages, NK cells, granulocytes, activated lymphocytes, and B-1 cells in the peritoneal cavity. M1/70 also cross-reacts with human CD11b and can detect this antigen on human peripheral blood monocytes, granulocytes, and some NK cells. CD11b participates in adhesive cell interactions.

Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocyte or bone marrow cell suspensions. Recommended ≤0.25 µg per test (in 100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Optimize titration for best performance.

Fluorochrome Information:
Alexa Fluor® 700 emits at 723 nm; excitation with red laser (633 nm). Most instruments require a 685 LP mirror and 710/20 filter.

Target Information

CD11b (integrin alpha-M, ITGAM):
A 165 kDa adhesion molecule that associates with integrin beta-2 (CD18) to form the CD11b/CD18 complex (Mac-1, CR3). Expressed on monocytes/macrophages, granulocytes, activated lymphocytes, subsets of NK and dendritic cells, and microglia. Functions as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X, mediating cell-cell and cell-matrix interactions. Expression varies by cell type and tissue; used as a microglial marker in nervous tissue. CD11b dysfunction is linked to systemic lupus erythematosus and ITGAM-related susceptibility.
