Target Information

The tachykinins are a family of small peptides that include neurotransmitters such as substance P, neurokinin A, and neurokinin B. These act on three related but distinct seven-transmembrane G-protein-coupled receptors at varying concentrations. The NK-3 receptor (NK3R) has the highest affinity for neurokinin B and is highly expressed in the supraoptic and paraventricular nuclei. Upon ligand binding, NK3R activates a phosphatidylinositol-calcium second messenger system, likely leading to the release of vasopressin and oxytocin. NK3R may play roles in learning and memory, as knockout mice exhibit cognitive deficits. It may also influence vagal afferent relay neurons, although activation in airway nerves is more likely mediated by substance P or neurokinin A rather than neurokinin B.
