Product Specific Information
The RPA-T8 monoclonal antibody reacts with human CD8a, a 32–34 kDa cell surface receptor expressed as CD8 alpha/beta heterodimer or CD8 alpha/alpha homodimer. CD8a is found on most thymocytes, mature T cells, and NK cells. It binds MHC class I molecules and associates with protein tyrosine kinase p56lck, contributing to T-cell development and activation.

- Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.25 µg)/test
- Recommended Use: Final volume 100 µL; 10⁵–10⁸ cells/test
Brilliant Violet™ 605 (BV605) emits at 605 nm and is used with a violet (405 nm) laser. Ensure instrument compatibility.
Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.
Light Sensitivity:
BV605 is sensitive to photo-induced oxidation. Protect vial and samples from light.
Fixation Compatibility:
Samples may be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark. BV605 is not compatible with methanol-based fixation.

Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family, crucial for T-cell signal transduction, differentiation, and activation. CD8 acts as a co-receptor for MHC class I molecules, while CD4 binds MHC class II. CD8 is expressed on T cells, NK cells, monocytes, and dendritic cells, forming disulfide-linked dimers of CD8α and CD8β or CD8αα. MHC-I/peptide complex binding recruits Lck kinase, leading to lymphokine production and cytotoxic T lymphocyte (CTL) activation. Activated CTLs contribute to pathogen and tumor clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
