Product Specific Information

The M1/70 monoclonal antibody reacts with mouse CD11b (integrin alphaM, 165–170 kDa). CD11b associates with CD18 to form the alphaMbeta2 integrin (Mac-1), binding to CD54 (ICAM-1), C3bi, and fibrinogen. It is expressed on macrophages, NK cells, granulocytes, activated lymphocytes, and B-1 cells in the peritoneal cavity. M1/70 also cross-reacts with human CD11b, enabling detection on human monocytes, granulocytes, and NK cell subsets.

Fluorochrome Information:
Brilliant Violet™ 605 (BV605) emits at 605 nm, excitable by a 405 nm laser. Ensure instrument compatibility.

Staining Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect from light due to photo-induced oxidation sensitivity.

Fixation Compatibility:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark. BV605 is not compatible with methanol-based fixation.

Target Information

CD11b (integrin alpha-M, ITGAM) is a 165 kDa adhesion molecule forming a heterodimer with CD18 (integrin beta-2). The CD11b/CD18 complex (Mac-1, CR3) mediates adhesion via binding to ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X.
CD11b is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia. Expression increases with monocyte maturation and varies among macrophage subsets.
CD11b dysfunction is associated with systemic lupus erythematosus and ITGAM-related susceptibility to autoimmune disease.
