Product Specific Information

The eBioSN4 monoclonal antibody reacts with human CD9, a 24 kDa member of the tetraspanin family characterized by four hydrophobic transmembrane domains.
CD9 is expressed in platelets, eosinophils, basophils, pre-B cells, activated T cells, and neural cell lines. Its expression is associated with malignant phenotypes, including non-T cell acute lymphoblastic leukemia and chronic lymphocytic leukemia.

In platelets, CD9 is localized in alpha-granules and interacts with integrin αIIb/βIII to mediate platelet aggregation. CD9 also promotes pre-B cell aggregation, Schwann cell adhesion, and migration. It can provide a co-stimulatory signal for T cells independently of CD28.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human platelets.
Use at 5 µL (0.06 µg) per test in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Target Information

CD9 antigen is a glycoprotein expressed on developing B lymphocytes, platelets, monocytes, eosinophils, basophils, stimulated T lymphocytes, neurons, and glial cells.
It belongs to the tetraspanin family, spanning the membrane four times. CD9 regulates cell activation and aggregation, possibly through association with integrin CD41/CD61 (GPIIb/GPIIIa).

CD9 plays roles in:

• Cell motility, osteoclastogenesis, neurite outgrowth, myotube formation, and sperm-egg fusion
• Cell attachment and proliferation
• Association of MHC II molecules on dendritic cells for effective T cell stimulation
• Suppression of cancer cell motility and metastasis

CD9 is considered a metastasis suppressor in solid tumors.
