Immunohistochemistry (Paraffin) [IHC (P)]

  • Tested Dilution: 10 µg/mL
  • Recommended: Use heat-induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen.

Target Information

TNIK (TRAF2 and NCK Interacting Kinase) is characterized by an N-terminal kinase domain and a C-terminal GCK domain that serves a regulatory function. TNIK is mainly expressed in brain, heart, and spleen, and acts as a specific effector of RAP2, regulating the actin cytoskeleton.
It is autophosphorylated in a manner dependent upon Lys54 in the ATP-binding pocket of its kinase domain and plays a major role in cytoskeleton regulation. TNIK also phosphorylates gelsolin, an actin-severing protein, in vitro, suggesting its function in cytoskeletal regulation.
