Product Specific Information

The eBioC9B7W monoclonal antibody recognizes mouse CD223 (LAG-3, LAG3) protein expressed by activated α/β-TCR-bearing T cells, a subset of γ/δ-TCR-bearing T cells, and a subset of NK cells. CD223 is a 70 kDa type I transmembrane protein structurally similar to CD4. A soluble form of human CD223 has been detected in serum, and mouse CD223 is known to be proteolytically cleaved, generating a 54 kDa extracellular fragment and a 16 kDa intracellular fragment.

CD223 binds MHC class II with higher affinity than CD4 and is involved in the negative regulation of T-cell activation and homeostatic proliferation. It is also expressed by CD4⁺CD25⁺ regulatory T cells and may contribute to their suppressive function.

Applications Reported: Flow cytometry, immunoprecipitation, ELISA, and functional blocking (in vitro/in vivo).
Applications Tested: Tested by flow cytometric analysis of anti-CD3 and anti-CD28-activated mouse splenocytes. Recommended ≤0.5 µg/test (100 µL final volume). Optimize concentration for each assay.

Target Information

LAG-3 is a 70 kDa surface glycoprotein of the Ig superfamily, homologous to CD4. It binds to MHC class II with higher affinity than CD4 and participates in the negative regulation of T-cell activation and proliferation. LAG-3 is expressed on activated T cells (including regulatory T cells) and NK cells, with higher expression on CD8⁺ T cells than CD4⁺ T cells. Coexpression of LAG-3 and CD49b marks Type 1 regulatory T cells (Tr1) in humans and mice.
