Product Specific Information

The monoclonal antibody eBio440c recognizes Siglec-H, a protein exclusively found on plasmacytoid dendritic cells (pDCs) or type I IFN-producing cells (IPC) in naive mice.
Mouse IPCs are typically PDCA+, CD11c+, CD11b-, B220+, and Ly-6C+, and respond rapidly to viral infections.

Siglec-H is a transmembrane protein of the Ig superfamily that binds sialic acid but lacks the cytoplasmic ITIM domain. It associates with DAP12 to enable signal transduction.
This antibody has been shown to inhibit pDC function (reduces IFNα secretion in response to CpG).

Some mouse strains (e.g., SJL) have higher pDC percentages compared to C57BL/6.

Applications Tested:

• Tested on mouse splenocytes
• Recommended ≤ 0.125 µg/test (100 µL final volume)
• Cell number: 10⁵–10⁸ cells/test
• Optimize titration for best results
