Product Specific Information

Description:
The IM7 monoclonal antibody reacts with all isoforms of mouse CD44 (Pgp-1). CD44 is expressed by hematopoietic and non-hematopoietic cells. Bone marrow myeloid cells and memory T cells highly express this antigen, and peripheral B and T cells can upregulate CD44 expression. CD44 functions as an adhesion molecule through its binding to hyaluronate, an extracellular matrix component.

Applications Reported:
This IM7 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.5 µg per test (100 µL volume, 10⁵–10⁸ cells/test). Optimize antibody titration for best performance.

Fluorochrome Information:
Brilliant Ultra Violet™ 615 (BUV615) is a tandem dye emitting at 615 nm, designed for use with UV (355 nm) lasers. Ensure instrument compatibility.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness.

Target Information

CD44 is a 100 kDa type 1 transmembrane glycoprotein expressed on human leukocytes, brain white matter, and epithelial cells of the intestine and breast. It acts as a receptor for hyaluronic acid (HA) and mediates cell-cell interactions, adhesion, and migration. CD44 participates in lymphocyte activation, recirculation, and homing. Expression may increase in certain carcinomas, potentially linked to metastasis. Alternative splicing generates multiple isoforms contributing to structural and functional diversity. Diseases associated with CD44 dysfunction include superficial keratitis and lichen sclerosus.
