Preparation:
Produced from sera of goats immunized with highly pure Recombinant Murine TNF-alpha. The anti-Murine TNF-alpha-specific antibody was purified by affinity chromatography using an immobilized Murine TNF-alpha matrix.
Sandwich ELISA:
To detect Murine TNF-alpha by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL is required. This antibody, used with PeproTech Biotinylated Anti-Murine TNF-alpha (500-P64GBt) as detection antibody, enables detection of at least 0.2–0.4 ng/well of Recombinant Murine TNF-alpha.
Western Blot:
For detection by Western blot, use at 0.1–0.2 µg/mL. When combined with compatible secondary reagents, the detection limit for Recombinant Murine TNF-alpha is 1.5–3.0 ng/lane under reducing or non-reducing conditions.
Target Information
TNF alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR receptors. TNF alpha regulates immune cell activity, proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It exists as multimers (dimers, trimers, or pentamers) and shows a single 17 kDa band in SDS-PAGE under non-reducing conditions. Knockout mouse studies suggest TNF alpha’s neuroprotective role, and it is known to induce tumor necrosis in tumor-bearing mice. TNF alpha is also involved in immune responses to bacterial, viral, parasitic, and fungal infections, and plays a role in tumor cytotoxicity synergistic with interferon-gamma.
This cytokine has been implicated in autoimmune diseases, insulin resistance, and cancer.
