Product Specific Information
The monoclonal antibody 20Raj1 recognizes the human Ki-67 protein. Two isoforms of Ki-67 exist (345 and 395 kDa) and are expressed in dividing cells. Ki-67 is present in all cell types during active cell cycle phases (G1, S, G2, and mitosis) but absent in resting cells (G0). During interphase, Ki-67 localizes to the nucleus and redistributes to chromosomes during mitosis, associating with heterochromatin-bound proteins such as CBX3.
In tumor studies, Ki-67 expression serves as a marker to determine the fraction of proliferating cells within a tumor population. This antibody also recognizes canine Ki-67.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) and protocol.

Target Information
Ki-67 is a nuclear protein expressed during G1, S, G2, and M phases of the cell cycle. It contains a forkhead-associated (FHA) domain that associates with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus. Ki-67 shows phase-dependent localization: perinucleolar (G1), nuclear matrix (G2), and chromosomal (M phase).
Ki-67 is widely used as a proliferation marker since it is absent in G0 cells but increases from G1 through mitosis. It helps determine the growth fraction in neoplasms, correlating with tumor grade and prognosis. It is also used as a neuronal proliferation marker.
