Target Information
VEGF (vascular endothelial growth factor)는 45 kDa 크기의 이황화 결합된 호모다이머성 당단백질로, 신생혈관 형성(angiogenesis)에 관여하며 종양 성장과 전이에 중요한 역할을 합니다. VEGF는 혈관내피세포의 생존, 증식, 화학주성 및 투과성을 촉진합니다. VEGF 패밀리에는 VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E, PIGF가 포함됩니다. VEGF와 그 수용체 시스템은 혈관신생 세포 신호 전달의 핵심 조절자로 알려져 있습니다. 대부분의 종양은 혈관신생을 필수적으로 필요로 하며, VEGF는 가장 강력한 혈관신생 촉진 사이토카인으로 보고되었습니다. VEGF는 대체 스플라이싱을 통해 생성된 5가지 주요 isoform을 가지며, 각각은 tyrosine kinase 수용체 FLT1(VEGFR-1)과 FLK1/KDR(VEGFR-2)에 결합합니다. 이 수용체들은 주로 내피세포에서 발현됩니다. FLT1의 대체 스플라이싱 형태인 sFLT1은 VEGF를 높은 친화도로 결합하여 혈관신생 억제 역할을 합니다. VEGF의 상승된 수준은 POEMS 증후군(Polyneuropathy, Organomegaly, Endocrinopathy, Monoclonal gammopathy, Skin changes)과 관련이 있습니다.
