Product Specific Information
Description:
The eBioH4A3 monoclonal antibody reacts with human CD107a (LAMP-1), a highly glycosylated 110 kDa protein mainly localized in lysosomal/endosomal membranes. CD107a transiently appears on the surface of degranulating cytolytic T cells and is upregulated on activated platelets and certain cancer cells.
Applications Reported:
- Suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested:
- Pre-titrated and validated on Jurkat cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
- Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Fluorochrome Information:
- PerCP-eFluor® 710 emits at 710 nm and is excited at 488 nm (blue laser).
- Recommended filter: 710/50 bandpass (695/40 acceptable).
- Light-sensitive: protect from light.
- Fixation: Compatible with IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333); stable up to 3 days at 4°C in dark.
- Filtration: 0.2 µm post-manufacturing filtered.
Target Information
LAMP1 (CD107a)
LAMP1, together with LAMP2, is a major component of lysosomal membranes. About 1–2% is also found on the plasma membrane. It is a heavily glycosylated type I integral membrane protein involved in lysosome biogenesis and fusion with phagosomes. Upon cell activation, LAMP1 translocates to the plasma membrane via a tyrosine-based motif (YXXI).
LAMP1 promotes adhesion of PBMCs to vascular endothelium and is linked to inflammatory responses. Increased expression is noted in neurons and glial cells near senile plaques in Alzheimer’s disease and in immune cells during thymic involution. LAMP1 serves as a reliable marker for mast cell activation.
