Target Information
Calcineurin is a Ser/Thr, calcium and calmodulin-dependent protein phosphatase that plays an essential role in the T cell activation pathway. It consists of two subunits:
- Calcineurin A (CnA): 60 kDa catalytic subunit, homologous to protein phosphatases 1 and 2A
- Calcineurin B (CnB): 19 kDa regulatory subunit containing four EF-hand calcium-binding sites
Calcineurin-mediated dephosphorylation of NF-AT is essential for its activation, nuclear translocation, and gene expression in T-cells. Immunosuppressive drugs such as cyclosporin A and FK506 inhibit NF-AT activation by forming complexes with immunophilins that block substrate access to the catalytic domain of CnA.
