Target Information
TLR4는 선천면역 활성화와 병원체 인식에 관여하는 Toll like receptor (TLR) 패밀리의 구성원입니다. 이 수용체는 태반과 백혈구의 myelomonocytic 아형에서 가장 풍부하게 발현됩니다. 포유류 세포는 LPS에 반응하여 TLR4를 활성화하며, CD14, LY96, TLR4로 구성된 다단백질 복합체의 일부로 작용합니다. TLR4는 병원체 관련 분자 패턴(PAMPs)을 인식하고, 면역 반응에 필요한 사이토카인 생성을 매개합니다. TLR4 유전자 변이는 LPS 반응성 차이와 연관되며, 여러 전사 변이체가 보고되었습니다. TLR4는 말초혈 단핵구와 일부 B세포, 인간 태반에서 발현됩니다. TLR4 결핍 생쥐는 LPS에 대한 반응이 결여되어 그람음성 패혈증에 대한 감수성이 증가합니다.
