Product Specific Information

This MIH64 monoclonal antibody recognizes mouse VISTA, also known as B7-H5 or PD-H1.
It is validated for flow cytometry on both native and paraformaldehyde-fixed cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes, ≤1 µg/mL per test (100 µL final volume).
Cell number may vary between 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal results.

Super Bright 436 is excited by the violet laser line (405 nm) and emits at 436 nm.
Recommended filter: 450/50 bandpass or equivalent.
Ensure instrument compatibility with this fluorochrome.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

VISTA (V-domain Ig containing suppressor of T-cell activation), also known as B7-H5 or PD-1H, is encoded by the VSIR gene.
Although homologous to PD-L1, VISTA has a distinct expression pattern—predominantly on hematopoietic cells, especially myeloid cells, and at lower levels on T cells. It is typically absent on B cells.
VISTA expressed by antigen-presenting cells can directly suppress CD4 and CD8 T-cell proliferation and cytokine production, making it a promising target for cancer immunotherapy.
Reported ligands include CD28H, VSIG-3, and VSIG-8.
