Target Information

Gamma-aminobutyric acid (GABA) is the primary inhibitory neurotransmitter in the central nervous system, causing membrane hyperpolarization through the opening of a Cl⁻ channel associated with the GABA-A Receptor (GABA-A R) subtype. GABA-A Rs are crucial therapeutic targets for sedative, anxiolytic, and hypnotic agents, and are implicated in diseases such as epilepsy, anxiety, depression, and substance abuse.

The GABA-A R is a multimeric subunit complex with six alpha, four beta, and four gamma subunits identified, along with splice variants. Expression of alpha and beta subunits results in functional GABA-A Rs sensitive to GABA, while coexpression of a gamma subunit is required for benzodiazepine modulation. Benzodiazepine effects in the brain may be mediated by different alpha subunits, and phosphorylation of beta subunits can modulate receptor function.
