Product Specific Information

The NP-6G4 monoclonal antibody reacts with human CD195 (CCR5), an approximately 45 kDa member of the G protein-coupled receptor (GPCR) superfamily that binds chemokines including MIP-1α, MIP-1β, and RANTES.
Human CCR5 is expressed on resting T cells with a memory/effector phenotype, monocytes, macrophages, and immature dendritic cells. CCR5 functions as a receptor to direct lymphocyte chemotaxis and serves as the primary co-receptor for macrophage-tropic HIV-1, which binds CCR5 through gp120.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells.
For optimal performance, stain at 37°C for 30–60 minutes in Flow Cytometry Staining Buffer.
Use 5 µL (0.25 µg) per test (100 µL total volume). Cell number range: 10⁵–10⁸ cells/test.
Excitation: 633–647 nm; Emission: 660 nm; Laser: Red Laser.
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

HIV and related viruses require coreceptors, in addition to CD4, to infect target cells. Several chemokine receptors such as CCR5, CXCR4, CCR3, CCR2b, and CCR8 function as HIV coreceptors. Among them, CCR5 (CC-CKR-5) is the principal co-receptor for macrophage- and dual-tropic HIV-1 strains, mediating fusion and entry into human white blood cells.
CCR5 is essential for infection by HIV-1, HIV-2, and SIV. Its ligands—RANTES, MIP-α, and MIP-β—can prevent infection by M-tropic HIV-1. CCR5 interacts with the CD4-gp120 complex on HIV, leading to membrane fusion and viral entry. The amino-terminal domain and extracellular loops of CCR5 serve as HIV binding sites. CCR5 mRNA is expressed in lymphoid organs and monocytes.
