Target Information

PTEN is a tumor suppressor frequently mutated in various cancers. It encodes a phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase with tensin-like and catalytic domains. Unlike typical protein tyrosine phosphatases, PTEN preferentially dephosphorylates phosphoinositide substrates, negatively regulating intracellular phosphatidylinositol-3,4,5-trisphosphate levels and the AKT/PKB signaling pathway. PTEN modulates AKT-mTOR signaling, influencing neuron integration, positioning, dendritic development, and synapse formation in adult neurogenesis. It may also negatively regulate insulin signaling and glucose metabolism in adipose tissue. The nuclear monoubiquitinated form has greater apoptotic potential, while the cytoplasmic nonubiquitinated form shows reduced tumor suppressive ability.
