Target Information

Insulin and Insulin-like Growth Factor receptors are transmembrane tyrosine kinase receptors that play critical roles in development, cell growth, and metabolism. Binding of insulin or IGF-1 to the extracellular domain of IR and IGFR leads to receptor autophosphorylation and activation of intrinsic tyrosine kinase activity, enabling phosphorylation of appropriate substrates.

Phosphorylation sites unique to each receptor contribute to distinct signaling mechanisms. The catalytic loops within the tyrosine kinase domains contain motifs such as Tyr1158, 1162, and 1163 (IR) and Tyr1131, 1135, and 1136 (IGF1R). Autophosphorylation proceeds sequentially, initiating at the second tyrosine (1162 or 1135), followed by phosphorylation at 1158 (IGF-1R 1131) and 1163 (IGF-1R 1136), resulting in full activation of IR and IGF1R.
