Target Information
RNF168 is a chromatin-associated RING finger protein functioning as a ubiquitin ligase both in vitro and in vivo. It targets histones H2A and H2AX to form K63 polyubiquitin chains. Upon DNA double strand breaks, RNF168 localizes to damage sites in an RNF8-dependent manner, co-localizing with γH2AX and 53BP1. This promotes retention of 53BP1 and BRCA1, facilitating downstream DNA damage response signaling. RNF168 thus plays a key role in genome maintenance, with at least three known isoforms.
