Description:
The monoclonal antibody 3D12 (HLA-E) recognizes human HLA-E, a member of the Human Leukocyte Antigen family. HLA-E is a MHC Class IB molecule involved in NK cell recognition. It binds a restricted set of peptides from other Class I leader sequences to form a complex with β2-microglobulin (B2M), which interacts with NKG2A on NK cells, protecting HLA-G-expressing cells from NK cell-mediated killing. HLA-E expression is ubiquitous.

Reported for use in flow cytometric analysis.
Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use ≤0.5 µg per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance.

Target Information
HLA-E is a class I heavy chain paralogue forming a heterodimer with β2-microglobulin. It binds a restricted subset of peptides derived from leader sequences of other class I molecules. The heavy chain (~45 kDa) is membrane-anchored and encoded by 8 exons:
- Exon 1: Leader peptide
- Exons 2–3: α1, α2 domains (peptide-binding)
- Exon 4: α3 domain
- Exon 5: Transmembrane region
- Exons 6–7: Cytoplasmic tail
HLA and MHC antibodies are crucial in Immunopeptidomics, aiding neoantigen identification and characterization via HPLC-MS/MS.
