This antibody detects endogenous levels of ACK1 only when phosphorylated at Tyr859 and Tyr860, which have been historically referenced as Tyr857 and Tyr858.
Target Information
TNK2 (ACK1), also known as activated Cdc42-associated kinase, is a non-receptor tyrosine kinase that functions downstream of Cdc42. It plays a critical role in:
- Tumorigenesis and metastasis, particularly in prostate cancer
- CDC42-dependent cell migration via phosphorylation of BCAR1
- Synaptic function, brain development, and plasticity
Functional roles include:
- Activates AKT1 by phosphorylation on Tyr-176
- Phosphorylates AR on Tyr-267 and Tyr-363, promoting recruitment to androgen-responsive enhancers (AREs)
- Phosphorylates WWOX on Tyr-287
- Enhances MCF2 activity as a GEF toward Rho family proteins
- Regulates AXL receptor levels and contributes to metastatic potential by modulating tumor suppressors and pro-survival factors
