Target Information

The MYH9 gene, located on chromosome 22q12.3, encodes the heavy chain of non-muscle myosin IIA (NMHC IIA), a key component of the actin cytoskeleton that regulates essential cellular processes such as migration, adhesion, division, and shape maintenance.
The MYH9 gene spans over 106 kilobases with 41 exons encoding a 1,960-amino-acid protein forming a hexameric complex (two heavy chains, two regulatory light chains, and two essential light chains).
Mutations in MYH9 cause autosomal dominant MYH9-related diseases (MYH9-RD) such as May-Hegglin anomaly, Fechtner syndrome, and Epstein syndrome. These disorders are characterized by macrothrombocytopenia and can lead to hearing loss, renal failure, and cataracts.
MYH9 also plays a crucial role in hematopoiesis, being necessary for the survival and maintenance of hematopoietic stem and progenitor cells (HSPCs). Loss of MYH9 function disrupts blood cell formation, leading to severe deficiencies and bone marrow failure.
