Target Information
AIRE is a transcriptional regulator protein that binds DNA as a dimer and tetramer, but not as a monomer, and may regulate gene expression. It is a nuclear protein of 545 amino acids containing an HSR domain, two PHD-type zinc fingers, and a SAND domain. AIRE may form homodimers, homotetramers, and nuclear bodies, and interacts with transcriptional coactivators CBP and CREBBP. Three isoforms of AIRE are reported, one potentially involved in nuclear-mediated decay (NMD). AIRE is essential for immune regulation and exhibits E3 ubiquitin ligase activity. Mutations in AIRE cause the rare autosomal-recessive autoimmune disorder known as autoimmune polyendocrinopathy-candidiasis-ectodermal dystrophy (APECED).
