Product Specific Information

The PK136 monoclonal antibody reacts with mouse NK1.1, an antigen expressed by natural killer (NK) cells and a subset of T cells in NK1.1-positive mouse strains (e.g., C57BL and NZB). Common strains such as BALB/c, SJL, AKR, CBA, C3H, and A do not express NK1.1. For these strains, use the DX5 monoclonal antibody (Product #14-5971).

Simultaneous staining of C57BL/6 spleen cells with PK136 and DX5 reveals coexpression of both markers by most cells, with minor DX5⁺PK136⁻ and DX5⁻PK136⁺ subpopulations.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes at ≤0.25 µg/test.
A test is defined as the antibody amount staining a cell sample in 100 µL volume. Cell number may range from 10⁵–10⁸ cells/test. Titrate for optimal performance.

Light Sensitivity: PE-Cyanine7 is sensitive to photo-induced oxidation; protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information

NK1.1 (KLRB1, NKR-P1A, CD161) is a C-type lectin-like receptor expressed on natural killer (NK) cells and some T lymphocytes. It plays a role in NK cell cytotoxicity and cytokine secretion. The KLRB1 protein contains extracellular C-type lectin motifs, a transmembrane domain, and a cytoplasmic domain. It is a type II membrane protein with an external C-terminus. NKR-P1A recognizes LLT1 (Lectin Like Transcript-1) as its ligand.
