Target Information

Ubiquitin exists either covalently attached to another protein or free (unanchored). When covalently bound, it is conjugated to target proteins via an isopeptide bond either as a monomer (monoubiquitin), a polymer linked via different Lys residues of ubiquitin (polyubiquitin chains), or a linear polymer linked via the initiator Met of ubiquitin (linear polyubiquitin chains).
Polyubiquitin chains have different functions depending on the Lys residue linkage:

• Lys-6-linked: DNA repair
• Lys-11-linked: ERAD and cell-cycle regulation
• Lys-29-linked: lysosomal degradation
• Lys-33-linked: kinase modification
• Lys-48-linked: proteasomal degradation
• Lys-63-linked: endocytosis, DNA-damage response, NF-kappa-B signaling
  Linear polymer chains formed via Met linkage are involved in cell signaling.
  Ubiquitin is usually conjugated to Lys residues of target proteins, occasionally to Cys or Ser residues. Free polyubiquitin also plays roles in activation of protein kinases and signaling.
  60S ribosomal protein L40 is a component of the 60S ribosomal subunit, essential for translation of specific cellular transcripts, particularly cap-dependent translation of vesicular stomatitis virus mRNAs.
