Product Specific Information

The AER-37 monoclonal antibody reacts with the Fc epsilon RI alpha subunit, an IgE-binding subunit lacking signal-transducing ability. FcεRIα is expressed on mast and basophil cells and is upregulated by the presence of IgE. The FcεRIα forms a tetrameric complex with one beta and two gamma subunits, which contain immunoreceptor tyrosine-based activation motifs (ITAMs). This complex plays a key role in IgE-mediated allergic reactions.

• Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.5 µg) per test in 100 µL final volume.
• Excitation/Emission: 405 nm / 780 nm (Violet Laser)
• Recommended Filter: 780/60 bandpass

Super Bright 780 is a tandem dye excited by the violet laser line (405 nm) with emission at 780 nm. Protect from light as the dye is sensitive to photo-induced oxidation.

Compensation and Staining Notes

• When using UltraComp eBeads microspheres (Product # 01-2222-42) or AbC Total Antibody Compensation beads (Product # A10497), compensation values may differ from single-color stained cells. It is recommended to set compensation with cells.
• For panels using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Fixation

Samples can be stored in:

• IC Fixation Buffer ([Product # 00-8222-49])
• 1-Step Fix/Lyse Solution ([Product # 00-5333-57])
  for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.

Target Information

FceR1 (Fc epsilonR1 alpha) is a subunit of the high-affinity IgE receptor. The receptor complex consists of one alpha, one beta, and two gamma subunits. The beta and gamma subunits mediate signal transduction. FcεRI plays a central role in allergic responses by linking allergen-bound IgE to mast cells, triggering release of histamine and other mediators responsible for allergy symptoms such as those in hay fever and asthma.
