Specificity Comments:
Recognizes a cell surface glycoprotein of 95/115/135 kDa (depending upon glycosylation), identified as CD43 [Workshop IV].
70–90% of T-cell lymphomas and 22–37% of B-cell lymphomas express CD43. No reactivity with reactive B-cells.
Co-expression of CD43 in B-lineage cells suggests malignant lymphoma rather than reactive B-cell population.
Combined use with anti-CD20 enables effective immunophenotyping of lymphomas in formalin-fixed tissues.
Co-staining with anti-CD20 and anti-CD43 supports lymphoma diagnosis over reactive process.
Target Information
CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with high negative charge, expressed on most hematopoietic cells.
It contributes to a repulsive barrier interfering with adhesion but can promote leukocyte aggregation.
Through interaction with actin-binding proteins ezrin and moesin, CD43 regulates T-cell morphology and lymphocyte traffic.
CD43 signaling enhances LFA-1 adhesiveness and modulates its induction.
Expression of CD43 can induce tumor suppressor p53; however, in p53/ARF deficiency, it may promote tumor proliferation.
Associated diseases include Wiscott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
