• Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with high negative charge, expressed on most hematopoietic cells. It contributes to a repulsive barrier interfering with cellular adhesion but can also promote leukocyte aggregation. Through interactions with actin-binding proteins ezrin and moesin, CD43 regulates T-cell morphology and cell-cell interactions during lymphocyte traffic. CD43 signaling both enhances and counteracts LFA-1 adhesiveness depending on receptor context. Expression of CD43 can induce tumor suppressor p53, but in p53 or ARF deficiency, CD43 may promote tumor proliferation. CD43 is an important modulator of leukocyte functions.
Diseases associated with CD43 dysfunction include Wiscott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
