Product Specific Information

Description:
LN3 mAb reacts with human MHC class II molecule HLA-DR, expressed on antigen-presenting cells such as B cells, monocytes, macrophages, dendritic cells, and activated T cells. It plays a key role in peptide presentation to CD4⁺ T lymphocytes.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-diluted and tested on normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg)/test in 100 µL total staining volume. Cell count range: 10⁵–10⁸ cells/test.

Fluorochrome Information:
Brilliant Ultra Violet™ 805 (BUV805) emits at 797 nm and is designed for UV (355 nm) laser-equipped cytometers. Protect from light to avoid photo-induced oxidation.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer (Product # 00-4409-75) to minimize non-specific interactions.

Fixation Compatibility:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark. Compatible with short-term methanol-based fixation (≤1 hour).

Target Information

HLA-DR is a transmembrane glycoprotein composed of a 36 kDa alpha (DRA) and 27 kDa beta (DRB) chain. The DRA gene contains five exons encoding leader peptide, extracellular domains, transmembrane domain, and cytoplasmic tail. DRA acts as the sole alpha chain for DRB1, DRB3, DRB4, and DRB5. DRB1 allelic polymorphisms define peptide binding specificities, relevant for transplantation typing.

HLA-DR is primarily expressed on antigen-presenting cells (B cells, monocytes, macrophages, thymic epithelial cells, activated T cells). MHC class II molecules (DR, DQ, DP) bind processed peptides and present them to T-helper cells, initiating immune responses.

HLA and MHC antibodies are key tools in Immunopeptidomics, enabling identification and characterization of neoantigens via LC-MS/MS analysis.
