Product Specific Information

The monoclonal antibody B6H12 reacts with CD47, also known as integrin-associated protein (IAP) and neurophilin.
CD47 is a glycosylated five-transmembrane protein involved in cell adhesion through interactions with SIRP and integrins (CD51/CD61, CD41/CD61).
It mediates bi-directional signaling influencing neural synaptic activity and macrophage phagocytosis, and serves as a receptor for thrombospondin.
CD47 expression is found in most hematopoietic cells (T, B cells, monocytes, platelets, erythrocytes) and also in non-hematopoietic tissues.

This antibody has been reported to have neutralizing activity.

Applications Reported: Flow cytometry, immunoprecipitation, western blot, and immunohistochemistry (formalin-fixed paraffin-embedded sections).
Applications Tested:

• Flow cytometry: ≤1 µg/test (100 µL final volume, 10⁵–10⁸ cells/test)
• IHC (FFPE): ≤40 µg/mL using low pH antigen retrieval
  Optimal titration is recommended per assay.

Target Information

CD47 (Integrin-Associated Protein, IAP) is a ubiquitously expressed transmembrane glycoprotein that interacts with integrins and regulates multiple signaling pathways, including G protein activation and calcium signaling upon cell adhesion.
It functions as a receptor for the C-terminal domain of thrombospondin, playing roles in membrane transport and signal transduction.
CD47 binds to CD172a (SIRPα) on macrophages, inhibiting phagocytosis of CD47-positive cells.
Additionally, CD47-CD172a interactions influence cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development, being abundant in synapse-rich brain and retinal regions.
