SIRT3는 효모 Sir2 단백질과 상동성을 가지는 sirtuin 단백질 패밀리의 구성원입니다. sirtuin 패밀리는 sirtuin core domain을 특징으로 하며 네 가지 클래스로 분류됩니다. 인간 sirtuin의 기능은 아직 완전히 규명되지 않았으나, 효모의 sirtuin 단백질은 후성유전적 유전자 침묵 조절 및 rDNA 재조합 억제에 관여하는 것으로 알려져 있습니다. 연구에 따르면 인간 SIRT3 단백질은 mono-ADP-ribosyltransferase 활성을 가진 세포 내 조절 단백질로 작용할 수 있습니다. SIRT3는 sirtuin 패밀리의 class I에 속합니다.
