Product Specific Information

The CR55T33 monoclonal antibody recognizes phosphorylated serine 139 of human and mouse H2AX. H2AX is a member of the H2A histone family that complexes with DNA and other histones to form nucleosome units characteristic of eukaryotic chromatin. Nucleosomes consist of approximately 147 base pairs of DNA wrapped around an octamer of histones (H2A, H2B, H3, H4).

Upon DNA double-strand breaks caused by irradiation, genotoxic stress, replication errors, or recombination, PI3K-like kinases (ATM, ATR, DNA-PK) phosphorylate serine 139 on H2AX. This phosphorylation recruits DNA repair proteins.

The CR55T33 antibody detects a ~15 kDa band on reduced Jurkat cell lysates treated with etoposide. It is suitable for IHC (paraffin), ICC, and microscopy applications. Recommended use ≤10 µg/mL with optimization per assay.

Target Information

Histone H2A.X (H2AX) is a member of the histone H2A family, one of the four core histones forming the nucleosome core particle. DNA double-strand breaks trigger phosphorylation of serine 139 at the C-terminus of H2AX, forming γ-H2AX, detectable by Western blot or immunofluorescence.

Phosphorylation involves phosphatidylinositol 3-kinases such as ATM, DNA-PK, and ATR. H2A.X contains 143 amino acids and is synthesized in both G1 and S phases. Two mRNA forms exist: a short replication-type (575 bases) and a long basal-type (1600 bases). H2A.X maps to chromosome 11q23.2–q23.3 and regulates MDC1 binding at DNA damage sites.
