Product Specific Information

The 12G5 monoclonal antibody reacts with human CD184 (CXCR4), also known as Fusin, LESTR, or HUMSTR. CD184 is a G-protein-coupled chemokine receptor with seven membrane-spanning domains and functions as a coreceptor for X4 HIV-1 entry into CD4+ cells. It is predominantly expressed on naive T cells of peripheral blood and upregulated by PHA and IL-2 stimulation.

The 12G5 antibody partially inhibits chemotaxis and calcium influx induced by SDF-1, blocks CD4-independent HIV-2 infection, and blocks CD4-dependent infection by some T-tropic HIV-1 isolates.

Light Sensitivity: Tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark. Clone-specific performance should be determined empirically.

Target Information

CXCR4 (Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor that acts as a co-receptor for HIV-1 and HIV-2 entry via CD4. It binds CXCL12/SDF-1, mediating calcium signaling and MAPK activation. CXCR4 is involved in hematopoiesis, cardiac septum formation, and vascularization processes. In the CNS, it mediates neuron survival and cerebellar development. It also binds bacterial LPS and mediates inflammatory responses. Antibodies to CXCR4 block HIV-1 and HIV-2 fusion and infection.
