Product Specific Information

Description:
The W6/32 monoclonal antibody reacts with human MHC class I antigens (HLA-A, B, C). These antigens, associated with β2-microglobulin, are expressed by all nucleated human cells and are critical for cell-mediated immune responses and tumor surveillance. W6/32 recognizes a non-polymorphic epitope shared among HLA-A, B, and C loci and immunoprecipitates 43 kDa and 11–12 kDa chains. Cross-reactivity is observed in baboon, rhesus, and cynomolgus monkeys.

Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test (100 µL final volume). Optimal cell number: 10⁵–10⁸ cells/test.

Light Sensitivity:
Tandem dye is sensitive to photo-induced oxidation; protect from light.

Fixation:
Samples can be stored in:

• IC Fixation Buffer (Product # 00-822-49)
• 1-step Fix/Lyse Solution (Product # 00-5333-54)
  for up to 3 days at 4°C in the dark with minimal effect on brightness and FRET efficiency.

Target Information

HLA-class I MHC antigens are intrinsic membrane glycoproteins expressed on nucleated cells, noncovalently associated with β2-microglobulin. They present foreign peptides for recognition by cytotoxic T cells, playing a key role in antiviral and antitumor immune defense. Human HLA-class I antigens include HLA-A, HLA-B, and HLA-C molecules.

HLA and MHC antibodies are valuable in Immunopeptidomics, aiding in neoantigen identification and characterization via LC-MS/MS.
