Target Information
MERTK (cMER) is a tyrosine kinase proto-oncogene involved in retinal pigment epithelium (RPE) phagocytosis, which is associated with human retinal disease. It regulates cell survival, migration, differentiation, and efferocytosis (phagocytosis of apoptotic cells). Ligand binding induces autophosphorylation of MERTK, creating docking sites for downstream signaling molecules such as GRB2 and PLCG2, and triggers phosphorylation of MAPK1, MAPK2, FAK/PTK2, or RAC1. MERTK signaling contributes to macrophage clearance of apoptotic cells, platelet aggregation, cytoskeletal reorganization, and engulfment. In RPE, it regulates rod outer segment fragment phagocytosis and inhibits Toll-like receptor (TLR)-mediated innate immune responses by activating STAT1, which induces SOCS1 and SOCS3 expression.
