• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

MERTK (cMER) is a tyrosine kinase proto-oncogene involved in the retinal pigment epithelium (RPE) phagocytosis pathway, which is implicated in human retinal disease. It regulates cell survival, migration, differentiation, and efferocytosis. Ligand binding induces autophosphorylation of MERTK, creating docking sites for signaling molecules such as GRB2 and PLCG2, and activates MAPK1, MAPK2, FAK/PTK2, and RAC1. MERTK signaling contributes to macrophage clearance of apoptotic cells, platelet aggregation, cytoskeletal reorganization, and engulfment. In RPE cells, it regulates phagocytosis of rod outer segments. Additionally, MERTK inhibits Toll-like receptor (TLR)-mediated innate immune responses by activating STAT1, which induces SOCS1 and SOCS3 production.
