• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein that functions as a cell death receptor of the TNFR superfamily. It also exists in a soluble form (~26 kDa). Stimulation of FAS triggers aggregation of FAS-associated death domains (FADD), formation of the death-inducing signaling complex (DISC), and activation of caspases. This cascade leads to apoptosis. FAS can also activate NF-kappaB, MAPK3/ERK1, MAPK8/JNK, and is involved in proliferative signaling in fibroblasts and T cells. Multiple alternatively spliced transcript variants exist, some lacking the transmembrane domain and negatively regulating apoptosis. The Fas/Fas ligand system is implicated in diseases such as AIDS, hepatitis, and cancer, and plays a key role in antiviral immune responses.
