Product Specific Information

  • For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein functioning as a cell death receptor of the TNFR superfamily. It also exists in a soluble form (~26 kDa). Upon stimulation, FAS aggregates its intracellular FAS-associated death domains (FADD) to form the death-inducing signaling complex (DISC), activating caspases and triggering apoptosis.
FAS can also activate NF-kappaB, MAPK3/ERK1, and MAPK8/JNK pathways, playing roles in signal transduction in fibroblasts and T cells. Multiple alternatively spliced isoforms exist, some lacking the transmembrane domain and acting as negative regulators of apoptosis.
The Fas/Fas ligand system is implicated in diseases such as AIDS, hepatitis, and cancer, and plays a key role in antiviral immune responses.
