NAD-dependent protein deacetylase sirtuin-1 (SIRT1) links transcriptional regulation to intracellular energetics and coordinates several cellular processes including cell cycle, DNA damage response, metabolism, apoptosis, and autophagy.
SIRT1 modulates chromatin function through histone deacetylation and influences histone and DNA methylation, leading to transcriptional repression. It is essential in skeletal muscle cell differentiation and mediates inhibitory effects under low nutrient conditions involving AMPK and NAMPT.
