Product Specific Information

The monoclonal antibody 29A1.4 recognizes mouse NKp46 (CD335), a glycoprotein with two Ig-like domains and a short cytoplasmic tail. Expression of CD335 is unique to NK cells (including immature NK cells, DX5- CD3-), allowing discrimination between NKT and NK cells (NKp46+, CD3-).
Staining is not strain-specific and is shown on C57Bl/6, SJL, CBA/CA, and BALB/C strains. NKp46 plays a role in NK cell-mediated lysis of tumor and pathogen-infected cells.

The 29A1.4 antibody activates NK cells in vitro but does not deplete NK cells in vivo.

Applications Tested: Mouse splenocytes, ≤0.5 µg/test (100 µL final volume). Optimize titration for best performance.

Fluorophore Information:
Super Bright 600 is a tandem dye excited at 405 nm (violet laser) and emits at 600 nm. Recommended filter: 610/20 bandpass. Ensure instrument compatibility.

Staining Panels:
When using multiple Super Bright conjugates, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect vial and stained samples from light.

Fixation:
Samples may be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.
